I always get a laugh out of the scene in Columbo Goes to College when he's interviewing Dominic Doyle. Columbo says that he's from homicide and Doyle replies "I haven't done one of them lately you know?"

From The Conspirators (this one I had to look up):

Columbo on observing a flat tire: "Will you look at that? How do you like that? That's practically a brand new recap."

Devlin: "Can I give you a hand with the spare?"

Columbo: "Well, the spare, sir, that's back in my garage."

Devlin: "Very sensible. If you get a flat at home, the spare's out and ready."

Grants post made me realize that we could have a whole thread on Columbo's car just by itself. For instance, in A Friend in Deed. when he goes to see Charlie Shoop that interaction is hilarious. Shoop wants to know if there's 20 or 30,000 miles on the car. And then at the end of the conversation, he said that he would give him $200 for it in a trade. Hilarious stuff
